Quantitative In Vivo Assessment of Tumour Vasculature- Targeted Liposomes
نویسنده
چکیده
Targeting angiogenic vasculature has been validated as a viable approach for cancer imaging and therapy. The tumour vasculature-specific ligand asparagine-glycinearginine (NGR) peptide targets the isoform of aminopeptidase N (CD13) expressed on endothelial cells lining angiogenic vessels. CD13 has become widely recognized as a rational target for therapeutic development and several NGR-conjugated agents are now in pre-clinical and clinical development. In the current study, a CT image-based approach is used to evaluate the in vivo performance of several NGR-conjugated liposome formulations that vary in terms of NGR density and PEG spacer arm length. Indeed, for the first time it is demonstrated that CT imaging can be used for quantitative and longitudinal assessment of the pharmacokinetics and biodistribution of an actively targeted liposome formulation. In comparison to conventional methods, CT imaging enables visualization of the intratumoural distribution of liposomes and quantification of the fraction of tumour occupied by the vesicles over time.

متن کاملRGD-based strategies for selective delivery of therapeutics and imaging agents to the tumour vasculature.
During the past decade, RGD-peptides have become a popular tool for the targeting of drugs and imaging agents to alphavbeta3-integrin expressing tumour vasculature. RGD-peptides have been introduced by recombinant means into therapeutic proteins and viruses. Chemical means have been applied to couple RGD-peptides and RGD-mimetics to liposomes, polymers, peptides, small molecule drugs and radiot...
متن کاملIn vivo Delivery of Liposomal Encapsulated Survivin siRNA Leads to a Reduction in Tumour Growth Rate
Introduction The RNA interference (RNAi) mechanism has great potential in the treatment of cancer due to targeted inhibition of upregulated genes in the tumour by small interfering RNAs (siRNA). However, delivery of siRNA’s to tumour cells has many obstacles in vivo such as degradation by enzymes in the blood and non-specific cell uptake. A possible solution to this is the use of liposomes, whi...
متن کاملLiposomal drug delivery in multimodal cancer therapy
Encapsulating cytostatics into lipid vesicles, i.e. liposomes, improves tumour drug accumulation and reduce adverse effects. Liposomal doxorubicin (DXR) has been used in the treatment of a variety of cancers and may also be suitable for combining with other treatment modalities. By modulating liposomal membranes, liposomes can be made ultrasound (US) sensitive releasing encapsulated drug in tum...
متن کاملMRI Characterisation of a Novel Transgenic Mouse Model of Neuroblastoma
Introduction Neuroblastoma is the most common extracranial childhood solid tumour, accounting for between 7-10% of paediatric cancers, and originates in peripheral nerve tissues (1). The proto-oncogene MYCN is amplified in 25% of high-risk neuroblastoma and is associated with an aggressive tumour phenotype, enhanced tumour angiogenesis and poor clinical prognosis (2). The Mycn oncoprotein is hi...
